透明质酸产生菌的诱变育种及发酵条件

摘    要:本文研究了透明质酸(HA)产生菌的诱变育种及摇瓶发酵条件。以兽疫链球菌(Streptococcuszoepidemiccus)NCT6180为出发菌种,在普通加血培养基上研究了HA的产生能力,用正交法研究了野生菌的最佳培养条件,通过单菌落分离得到野生高产菌株。再通过紫外和NTG诱变得到溶血性较弱的菌株,部分菌株比出发菌株产量提高一倍以上。在摇瓶条件下,PH值的控制及葡萄糖的加入均有利于HA的产生。

Abstract:rimary researches on the mutation breeding of Streptococcus zooepidemiccusand its shaker-flask fermentational conditions were carried out. Wild Streptococcus zOOepidemiccus NCTC 6180 was adopted, its ability of HA-producing wade examined by commonblood medium, and its optimum cultivation conditions were studied by orthogonal method.From the fermentational results of single colony, a high HA-producing strain was acquired, further more studies on its UV-mutation and NTG-mutation were conducted, and meanwhile some strains low-homoclasis were obtained; a number of those colonys canproduce 100% HA more than the original strain. The controlling of PH and the adding of glucose under the condition of shaker-flask are beneficial to the production of HA.
