摘    要:为了提高联合固氮菌的抗逆性,采用电融合技术,在小麦根际联合固氮菌(Klebsielaoxytoca)与抗逆性强的枯草芽孢杆菌(Bacilussubtilis)之间实行了原生质体融合,并从再生的融合子中筛选到多株具有特定性状的后代菌株。经形态观察、革兰氏染色、抗生素抗性、耐热性、抗菌活性、固氮活性以及淀粉酶试验等多项指标测定,数株融合子表现出双亲的耐热、抑菌、固氮等优良性状,具有潜在的应用价值。

Abstract:In order to improve stress resistance of the nitrosen fixing bacteria, interfamilial protoplast fusion between Klebsiella oxytoca , a gram negative, wheat associated nitrogen fixing bacteria, and Bacillus subtilis , a gram positive bacteria with the ability of secreting anti bacterial proteins, was performed by virtue of electrofusion technique. A number of fusants with specific properties were selected from the regenerated colonies. Through a series of bioassay including morphological observation, gram dyeing, antibiotics resistance, heat tolerance, anti bacterial activity, nitrogen activity and amylase test, the results showed that several fusants exhibited the desirable characteristics of both the parental strains, such as heat resistance, pathogen inhibition and nitrogen fixation. Therefore these recombinant fusants have potential values for agricultural application.
