江西龙宫洞不同季节土壤微生物群落结构比较

摘    要:采用非培养法和传统培养法相结合,利用Illumina 高通量测序技术对江西省九江市彭泽县龙宫洞微生物的16S rDNA 和ITS相关区域进行测序及生物信息学分析,通过实时荧光定量PCR(qPCR)对洞穴土壤中细菌和真菌含量进行绝对定量,探究不同季节对洞穴土壤微生物群落结构的影响,同时观察相应可培养微生物分布情况.结果表明:随着季节的变化,洞穴土壤的理化性质发生改变,继而引起土壤菌群结构的改变,其中土壤含水率是影响菌群结构的重要因素之一; 利用高通量测序技术获得非培养微生物的群落分布,根据其基因组信息,调整培养条件将可能有助于实现未培养微生物的可培养化.

Abstract:A combination of non-culture method and traditional culture method is used,and Illumina high-throughput sequencing technology is utilized to sequencing and bioinformatics analysis for the microbial 16S rDNA and ITS-related regions in Longgong cave in Jiangxi Province.The content of bacteria and fungi in the soil is quantified absolutely by real-time fluorescent quantitative PCR(qPCR).To explore the effects of different seasons on the structure of the soil microbial community in the cave,the distribution of the corresponding cultivable microorganisms is observed.The results show that with the change of seasons,the physical and chemical properties of the cave soil change to some extent,which leads to the change of soil microbial structure,and the soil water content affects one of the important factors of the flora structure.High-throughput sequencing technology is used to get the community distribution of non-cultured microorganisms.And adjusting the culture conditions according to their genomic information may help to achieve culturability of uncultured microorganisms.
