高分子微球固定化脂肪酶的合成研究

摘    要:本实验采用悬浮聚合法制备苯乙烯-二乙烯苯-马来酸酐(St—DVB—cBA)三元共聚高分子微球(0.26~0.33mm),并用其作为脂肪酶的载体,进行脂肪酶的固定化。实验结果表明,制备固定化脂肪酶的相对最佳条件:脂肪酶加入量2.0mg,载体50mg,反应温度30℃E,反应时间6h,pH=7.0,最高固载率可达80.02%,酶活4150U/g载体。所制得的酶最佳催化条件:反应温度40℃,pH=7.38。

Abstract:A poly(St-DVB-CBA) microsphere material was synthesized by copolymerization of styrene(St) ,divinylbenzene(DVB) and cis -butenedinic(CBA). The composite microspheres have proportionable diameter(0. 26 -0. 33 mm), which were used as carrier by covalent bond for the process of immobilization of lipase. The experimental results showed that the obtained optimal condition for preparation immobilized lipase are as follows : enzyme load 2.0 mg, carrier 50 mg, immobilization time 6 h, immobilization temperature 30 ℃, pH = 7. O. The maximum binding efficiency reached 80.02% , the activity Of immobilized lipase was 4150 U/g carrier. The optimal temperature of immobilized lipase was 40 ℃ and the optimum pH of immobilized lipase was 7.38. Compared with free enzyme, the thermal and pH stabilities of the enzyme were increased after the immobilization.
