HPLC法测定中江丹参植株不同部位丹参酮Ⅰ、丹参酮Ⅱ的含量

摘    要:目的:建立高效液相色谱法同时测定丹参植株中不同部位丹参酮Ⅰ、丹参酮Ⅱ两种脂溶性成分的含量。方法:采DiamonsilTM C18色谱柱(250 mm×4.6 mm,5μm),流动相:甲醇-水(84:16),柱温:35℃,流速为1.0 mL/min,检测波长268 nm。结果:在此色谱条件下2种成分可完全分离。丹参酮Ⅰ、丹参酮Ⅱ的线性范围分别为0.000 51~0.10μg(r=0.999 9),0.002 9~0.058μg(r=0.999 7)。平均回收率丹参酮Ⅰ为98.7%(RSD为1.2%),丹参酮Ⅱ为100.2%(RSD为1.7%)。实验结果可为中江丹参植株地上部位的开发利用提供科学依据。

Abstract:To establish an HPLC for simultaneous determination of two lipid-soluble compounds: Tanshinone Ⅰ , Tanshinone lq in different parts of salvia miltiorrhiza of Zhongjiang. Methods: The column was Diamonsil TM C18 (250 mm× 4. 6 mm,5μm). The mobile phase consisted of a mixture of methanolwater(84 : 16), the column temperature was 35℃ ,the flow rate was 1. 0 mL/rain,the detection wave- length was set up at 268 nm. Results: The linearity was obtained over the ranges of 0. 000 51 - 0.10 p,g ( r =0. 999 9) for Tanshinone Ⅰwith average recovery of 98.7% ( RSD was 1. 2% ), 0.002 9 - 0.058 μg ( r = 0. 999 7) for Tanshinone Ⅱ with average recovery of 100.2% ( RSD was 1.7% ). Conclusion: The method is rapid, simple, and reliable, and can be used for simultaneous determination of two lipid-soluble compounds in different parts of salvia miltiorrhiza
