血蓝蛋白与IgSF同源关系的研究

摘    要:采用蛋白质序列比对、同源建模和分子进化树构建等生物信息学技术,探索凡纳滨对虾血蓝蛋白与免疫球蛋白超家族(immunoglobulin superfamily,IgSF)的同源关系.结果发现,血蓝蛋白75kDa单体Ig-like区与人Ig存在5个同源模序,同源性为52%~69%,其中IgLH611-625模序与其同源序列(IgH41-55)的模拟三维结构同样非常相似.进一步研究显示,IgLH611-625模序不仅与多种高等生物IgSF分子具有同源性,而且在系统进化树中可与5种IgSF分子归属于同一大类.提示,血蓝蛋白确实与IgSF分子具有同源性.

Abstract:In the paper, an attempt was made to investigate the homology between Litopenaeus vannamei hemocyanin and immunoglobulins superfamily(IgSF) molecules. The bioinformatics technologies including protein-protein BLAST, homologous modeling and phylogenetie tree construction were used. The results showed that hemocyanin shared 5 homologous motifs with human Ig, and their homologous rates were 52%-69%, in which, the 3D structure of IgLH611-625 motif was very similar to its homologous sequence(Homo sapiens IgH41-55). Furthermore, the IgLH611-625 motif not only shared high homology with many IgSF molecules from different animals, but also could be attributed to the same class in the phylogenetic tree with five IgSF molecules. Thus, these data could be deduced that hemocyanin indeed present homology with IgSF molecules.
