甲磺酸帕珠沙星荧光性质的研究

摘    要:测定了溶液酸度、缓冲溶液种类及其用量、溶剂和干扰物等对甲磺酸帕珠沙星(PZFX)荧光性质的影响,建立了简便快捷的荧光光度法测定PZFX的定量方法.实验表明,PZFX在pH=4.8的HAc-NaAc缓冲体系中荧光强度最大,且缓冲液用量对PZFX荧光强度影响不大.干扰试验表明该方法的抗干扰能力比较强.当λex/λem=252nm/402nm时,PZFX浓度在1.00×10-8~2.00×10-6 mol/L范围内,其荧光强度与其浓度呈良好的线性关系,相关系数r=0.9993,检出限4.00×10-10mol/L.当PZFX浓度为1.00×10-7 mol/L时,测得相对标准偏差为2.1%.该法可用于实际样品注射液中PZFX含量的测定.

Abstract:A simple and convenient fluorescent measurement method to quantitatively determine pazufloxacin mesilate(PZFX) was established.The fluorescence properties of PZFX in different kinds of acidity,buffer solutions and their dosages,solvents and interferents were investigated.The results showed that PZFX had the highest fluorescence intensity in HAc-NaAc buffer solution with pH=4.80.The dosages of buffer solutions had less influence on the fluorescence intensity of PZFX.The method has relatively high-resistance to interferents and thus can be used in a complex system where some interferents exist.Under the optimal conditions,the fluorescence intensity was in a good linear relationship with its concentration in the range of 1.00×10-8~2.00×10-6mol/L,at λex/λem=252nm/402nm.The relevant coefficient of its calibration curve was 0.9993.The detection limit was estimated to be 4.00×10-10mol/L for PZFX.The relative standard deviation was 2.1% for 1.00×10-7mol/L PZFX.The proposed method can be applied to determine the quantity of PZFX in an injection liquid with satisfactory results.
