摘    要:人输卵管上皮细胞(HOEC)可以体外较长时间无血清培养,DMEM/F12培养液的培养效果优于Ham’sF10,在加有多种因子和胎球蛋白的培养液中,HOEC可贴壁生长,生长能力达到合质量分数15%FCS的对照组的30%~40%,一般可传代2次,最长培养可维持38d,细胞形态与有血清培养差别不大,虽然无血清培养HOEC的生长速度、密度、传代成功率远不及有血清培养,但能抑制成纤维细胞生长,维持较纯的上皮细胞。

Abstract:Cultuze of OEC has always been reported in medium with high serum supplements. In this study, we found that HOEC may grow in serum-free medium and last quite a few days. Media based on two different hasal media formulations were used in this study: DMEM/F12 and Ham's F10, and we found that DMEM/F12 was better than F10. Addition of several factors and fetuin to the medium enhanced HOEC attached tO the bottom of culture dish and made the ability of growth reach 30% ~ 40% of control group (addition of 15% FCS to medium). HOEC could be maintained alive through 2 ~ 3 passages, the longest culture was maintained for 38 days and the morphology and behavior of these cells maintained normal. Although the growing speed, the concentration and the ability of successful passage of HOEC which cultured in serum- free media were lower than cultured with high serum supplements, but serum - free culture can inhibit the growth of fibroblasts and maintained a higher purify epithelial cells growth.
