| 摘 要： | 制备掺Ｕ模板是利用大肠杆菌菌株ＣＪ２３６（ｄｕｔ＾－ｕｎｇ＾－Ｆ’）中，缺少一种酶，不能去除ＤＮＡ中的尿嘧啶残基。在含有尿苷的培养液中，在ＣＪ２３６中培养适当的Ｍ１３噬菌体，制备模板ＤＮＡ，时行滴度检测，根据结果可知：掺Ｕ模板制备出来了，然后将ＤＮＡ抽提出来，进行１％琼脂糖凝胶紫外检测根据结果可知：抽提出的掺Ｕ模板纯度很好。

| Abstract: | Preparation of template incorporated U is due to lack a kind of enzyme (Uracil N-glycosidase),which makes it is impossible to remove Uracil residue of DNA.Train proper phage MB in remove Uracil in culture fluid including Uracil,prepair template DNA and make titration inspection.We know template incorporated U has been prepared.Then extract DNA and make one percent agarose gel ultraviolet inspection.The outcome shows the purity of extractive template incorporated U is bggd. |
