紫花苜蓿多糖提取工艺的优化

摘    要:选择热水浸提法作为苜蓿多糖(APS)的提取方法,以浸提时间、浸提温度、浸提固液比以及浸提次数作为单因素进行单因素试验,通过方差分析,并进行最小显著差异法(LSD)多重比较作显著性分析,确定其条件范围。再通过进一步的正交实验L9(34)得到热水浸提法优选因素组合。结果表明:在浸提时间1 h、浸提温度95℃、浸提固液比1∶30、浸提3次的条件下,经醇析、氯仿-正丁醇溶液(4∶1)分离纯化,得脱蛋白粗多糖,多糖的提取率可达6.1%。

Abstract:Water extraction method was selected for polysaccharides extraction from alfalfa. The ex- traction time, temperature, ratio of solid to liquid and extracting times were investigated. Through the analysis of variance and LSD multiple comparison for significant analysis, the conditions were determined. Optimized factor combination was determined by orthogonal experiment L9 (34) . The results show that the optimal combination of extraction is 1 h, 95 ℃, solid-liquid ratio 1 : 30 and three times.
