小鼠在体肺灌注制样方法体重依赖的灌注量的探讨

摘    要:为探讨小鼠在体肺灌注制样方法体重依赖的最适灌注量。通过采用在体肺灌注制样并根据体重从0至0.2ml/20g探索灌注量,固定后进行切片HE染色观察固定效果。结果表明在0.2ml/20g,肺组织灌注均匀,切片后组织结构清晰,人为假象较少。可见小鼠在体肺灌注制样灌注量为0.2ml/20g可取得良好的组织固定效果。

Abstract:In order to investigate the weight-dependent optimal perfusion volume of the fixation of murine lungs is in situ fixation before opening the thorax. Samples were prepared by fixation of murine lungs is in situ fixation before opening the thorax and the perfusion volume was probed according to the body weight from 0 to 0.2 ml/20 g. After fixing, the fixation effect was observed by sectioning HE staining. The results show that in 0.2ml/20g, the lung tissue was evenly perfused, the histological structure was clear after slicing, and there was less artificial artifact. It is concluded that in mice, 0.2ml/20g perfusion of the fixation of murine lungs is in situ fixation before opening the thorax can achieve good tissue fixation.
