金针菇菌丝体静置培养的研究

摘    要:在碳源为2%,氮源含量为0.1%,采用液本静置培养方法,研究了金针菇Fq-19菌株对碳源、氮源、矿质元素和生长因子的利用。试验表明,金针菇Fq-19菌丝营养对有机氮的利用比对无机氮强,有机氮中,其对复合氮素如酵母粉、蛋白胨、黄豆粉的比单一氮素如尿素、谷胺酰胺强,在无机氮中,其对铵态氮的利用较强硝态氨基本上不被利用。金针菇Fq-19菌株对碳素营养的利用较很广泛,在多糖中,对玉米粉的利用最好。其次是可溶性淀粉,对纤维素的利用较差;单糖 的葡萄糖,双糖中的红糖、白糖、蔗糖、麦芽糖是理想的碳源,在缺碳素营养、氮素营养的培养基上菌丝不生长。添加生长因子可促进菌丝的生长,以葡萄为碳源,硫酸铵为氮源,金针菇Fq-19菌株适宜C/N为17-24:1,最适宜C/N为20:1。当C/N比为20:1时,菌丝生长旺盛,对营养利用较为合理。除碳、氮源外,维生素B1,K,P,M是金针菇菌丝生长的必须因子。

Abstract:Carbon nutrition and nitrogen nutrition utilization of Fq-19 strain of Flammulina velutipes was studies in quiet liquit cultur with 2% carbon nutrition and 0.1% nitrogen nutrition.The results showed that organie nitrogens were better utilized that inorganic nitrogens by Flammulina velutipes mycelia Among organic nitrogen,complex nitrogens,such as yeast meal,peptone,soybean meal were better utilized than simple nitrogens,such as urea and glutamine by the mycelia.Among inorganie nitrogens,ammonium nitrogen was well utilized,but nitrate nitrogen was hardil utilized.Flammulina velutipes mycelia utilized carbon nutrition widely.Maize meal was the best utilized polysaccharide,secondly was soluile starch and the last was cellulese.Glucose were well utilized monosaccharides,while maltose and sucrose were well utilized disaccharides.The optimum C/N ratio for Flammulina velutipes mycelia growth in liquid culture was 17-24:1,when glucose and (NH 4) 2SO 4 were used as carbon nutrition and nitrogen nutrition reps..At the C/N ratio of 20:1,the mycelia of Flammulina velutipes yrew well and the nutrition of the substrate was better used.Nutritional ingredients and their adequate concentration in medium were investigated by orthogonal evices VB 1 and the elements K,P,Mg,S also were essential additives,in addition to carbon and nitrogen sources,for the mycelia growth.
