| 摘 要： | 构建AhAO2基因片段原核表达重组质粒pPROEX-Hta-AhAO2,表达质粒在E.coliBL21中诱导表达,获得His-AhAO2融合蛋白,采用亲和层析与电泳纯化His-AhAO2免疫,新西兰大白兔,获得抗His-AhAO2抗体,West-ern blot检测显示其只与His-AhAO2融合蛋白结合.实验成功制备了抗AhAO2特异抗体.

| Abstract: | The prokaryotic expression plasmid pPROEX-Hta-AhAO2 was constructed.The results showed that the recombinant pPROEX-Hta-AhAO2 plasmid expressed high amount of His-AhAO2 fusion protein in E.coli BL21.An anti-AhAO2 polyclonal antibody was obtained by immunizing the New Zealand rabbit with His-AhAO2 fusion protein purified by affinity chromatography and electrophoresis.The antibody of AhAO2 binds specifically to His-AhAO2 fusion protein fragment in western blot analysis. |
