迎春花无菌体系的建立以及茎段和叶片愈伤组织的诱导

摘    要:本文对迎春花无菌体系的建立和茎段、叶片愈伤组织的诱导进行了研究。结果表明:(1)在建立迎春花的无菌体系时,应该选择晴天去采集外植体为好;(2)带芽茎段和叶片的污染率最大,而不带芽茎段的污染率最小;(3)在超净工作台外消毒应先用饱和洗衣粉溶液浸泡10 min,然后用自来水冲洗2-3次,最后再用蒸馏水冲洗2-3次;在进行超净工作台内消毒时,叶片应该先用84消毒液(1%)浸泡5min,无菌水冲洗3次,75%酒精浸泡20s,无菌水冲洗3次,0.1%升汞浸泡8 min,无菌水冲洗3次,而茎段应该先用84消毒液(1%)浸泡10 min,无菌水冲洗3次,75%酒精浸泡30s,无菌水冲洗3次,0.1%升汞浸泡10min,无菌水冲洗3次;(4)在NAA为0.2 mg/L时,高浓度的6-BA有利于不定根的再生,而低浓度的6-BA则更利于愈伤组织的诱导;在NAA为0.2 mg/L时,高浓度的KT有利于愈伤组织的诱导,而低浓度的KT则有利于不定根的再生;(5)NAA更利于不定根诱导,也可诱导愈伤组织。而2,4-D只利于愈伤组织的诱导,不利于诱导不定根。

Abstract:This article studied the establishment of Jasminum nudiflorum Lindl aseptic system and the induction of Stem and leaf Callus. The result indicated that: ( 1 ) when estabhshing Jasminum nudiflorum Lindl aseptic system, explants gathering should be chose in the cloudless day; (2) The pollution rate of the stems with a bud and the leaves was largest, but the pollution rate of the sterns without a bud was smallest; (3) the disinfection should first use the saturated laundry powder solution to soak 10 minutes outside the ultra - clean table, then flushes 2 - 3 time with the running water, finally uses the distilled water to flush again 2 - 3 time; When carrying on to disinfect in the ultraclean table, the leaves should first use 84 disinfection hquid ( 1% ) to soak 5 minutes, the aseptic water flushes 3 times, 75% ethyl alcohol soak 20 seconds, the aseptic water flushes 3 times, 0.1% mercuric chloride soak 8 minutes, the aseptic water flushes 3 times, but for the stems, they should first use 84 disinfection hquid ( 1% ) to soak 10 minutes, the aseptic water flushes 3 times, 75% ethyl alcohol soak 30 seconds, the aseptic water flushes 3 times, 0.1% mercuric chloride soak 10 minutes, the aseptic water flushes 3 times; (4) when NAA is 0.2mg/L, high density 6 - BA was advantageous to adventitious mot regeneration, but low density 6 - BA then favored callus induction; (4) when NAA is 0.2 mg/L, high density KT is advantageous to callus induction, but low density KT then was advantageous to adventitious mot regeneration; (5) NAA favored adventitious mot induction, also may induced callus. But 2,4 - D only favored callus induction, which is disadvantageous to the adventitious mot induction.
