愈伤组织悬浮培养法研究长喙田菁(Sesbania rostrata)根、茎、叶的重金属抗性

摘    要:
通过悬浮培养长喙田菁(Sesbania rostrata)愈伤组织,分别研究长喙田菁根,茎,叶各器官对重金属胁迫的反应,并量化各器官的重金属抗性,研究结果表明,愈伤组织悬浮培养方法能够有效测定植物根,,戒骄戒躁地的重金属抗生,长喙田菁根愈伤组织对Pb,Zn,Cu和Cd的抗性远高于茎和叶愈伤组织,叶愈伤组织则对各重金属较为敏感,其抗性只有根的1/8-1/4,Pb对根,茎,叶的半抑制质量浓度分别为80,40和15mg/L,Zn则分别为80,30和10mg/L,Cu分别为20.8和4mg/L,Cd分别115,8和6mg/L。

Abstract:
Heavy metal tolerances of root, stem and leaf of Sesbania rostrata were investigated by using callus suspension culture method. The results showed that the method was effective for testing the heavy metal tolerance of respective root, stem and leaf of S. rostrata. The highest tolerances to Pb, Zn, Cu and Cd were observed in the root callus. The tolerances in leaf callus were only 1/8~1/4 of that in root callus. ED-50s of Pb for growth of root, stem and leaf callus were 80, 40 and 15 mg/L, respectively. Those of Zn, Cu and Cd were 80, 30 and 10 mg/L, 20, 8 and 4 mg/L, and 15, 8 and 6 mg/L, respectively.
