摘    要:本实验以黄瓜(CucumissativusL.)品种“LS-21”为实验材料,按如下培养基配方:MS培养基,附加6-BA(0.8~1.6)mg/l IAA0.2mg/l;不同浓度的6-BA分别与NAA及2,4-D配合,研究影响黄瓜组织培养的诸多因素,建立了黄瓜离体子叶高频不定芽再生系统。主要结果如下:黄瓜5-6日苗龄无菌苗子叶作外植体,愈伤组织生长旺盛,芽诱导率高;7-8天的外植体则生长势弱,不利再生芽的分化。6-BA(1.2~1.6)mg/l IAA0.2mg/l,芽分化频率较高,其中6-BA1.2mg/l的芽分化率可达51%;待再生芽长至2-3cm时移入生根培养基(1/2MS 0.1mg/LIAA),生根率达90%,40-60d可得到完整再生植株。

Abstract:In this study,high efficiency of shoots regeneration conditions were established,and factors impacting cucumber tissue cultivation were studied by the basic medium MS with different hormones include 0.2 mg/L IAA and 0.8~1.6 mg/L 6-BA(W);6-BA with different concentration of NAA and 2,4-D for callus and shoots induction.The main results are as follows: callus growths well when use 5 day' cotyledons as explants than that of 7~8 days,the same results occurs in shoots regeneration rate.Higher efficiency of shoots regeneration obtained from MS medium added 0.2 mg/L IAA and 1.2~1.6 mg/L 6-BA(W),especially,in MS 1.2 mg/L 6-BA 0.2mg/l IAA,51% regeneration rate appeared,which is the highest rate of shoots regeneration in our experiments.The adventitious shoots were placed on medium(MS 0.1 mg/L IAA) for root regeneration,the rate is 90%.The whole regenerated plants could be obtained in 40-60 days.
