摘    要:应用RAPD技术对凉水国家自然保护区的天然红松(Pinus koraiensis)种群的遗传多样性变化和遗传分化进行了分析.共选择77个年龄在100~270年间的红松个体,连续分4龄级.筛选出11个随机引物共检测到71个位点,其中55个多态位点,多态位点比率为77.46%.利用POPGENE分析软件计算了该年龄段凉水地区红松遗传多样性指数,分析了遗传多样性在龄级间变化.研究结果表明,凉水保护区内的红松遗传多样性在1731-1900年的时间段内没有太大的波动;龄级间遗传多样性分化不明显;红松遗传多样性主要存在于龄级内.对照这一时期环境变化,对红松的遗传多样性演化与环境变化关系进行了讨论。

Abstract:RAPD (Random amplified polymorphic DNA) was applied to the study of the genetic diversity and differentiation in temporal scale of natural population of Pinus koraiensis forest in Liangshui National Nature Reserve. Total 77 P.koraiensis samples aged from 1731 to 1900 year were grouped into 4 age groups. Total 71 repeatable loci in which 55 were polymorphic have been checked using 11 random primers.The percentage of polymorphic loci (PPL) is 77.46%. With the POPGENE analysis software the genetic index of P.koraiensis in Liangshui National Nature Reserve was counted, and the differentiation of genetic diversity in age class level was analyzed. Result showed that genetic diversity of P.koraiensis in Liangshui National Nature Reserve didn't change too much during 1731 to 1900. In age class level the differentiation of genetic diversity is not obvious; and genetic diversity of P.koraiensis existed within age class. According to environment changes, the relationship between the evolvement of genetic diversity and environment changes was studied.
