辣根过氧化物酶在介孔碳CMK-3上在线固定化研究

摘    要:利用顺序注射-紫外可见分光系统,根据介孔材料孔径和生物大分子尺寸相当的特点,采用介孔碳CMK-3为载体实现对辣根过氧化物酶的在线固定化,并实时评价了固定化辣根过氧化物酶的活性及操作稳定性.紫外可见光谱和氮吸附实验结果表明:介孔碳CMK-3能有效吸附辣根过氧化物酶,理论固载量为54.95mg/g;辣根过氧化物酶不仅吸附在CMK-3表面,而且能进入其孔道内,从而增强固定化辣根过氧化物酶的活性稳定性.

Abstract:In the present work,on-line immobilization of horseradish peroxidase(HRP) on mesoporous carbon CMK-3 and real time evaluation of its activity have been researched by employing sequential injection-UV-Vis system.Comparing with the UV-visible spectroscopy of HRP before and after immobilization,it was found that HRP could be efficiently adsorbed by CMK-3 and the adsorption capacity was derived as 54.95 mg/g.The results of the N2 adsorption experiments showed that HRP was adsorbed not only on the surface of CMK-3 but also into the pore of CMK-3,which increased the operational stability of immobilized HRP.
