摘    要:植物半胱氨酸蛋白酶抑制剂是一类具有提高植物对各种生物及非生物胁迫抗逆性的重要蛋白质。利用前期获得的甜菜M14品系半胱氨酸蛋白酶抑制剂基因的编码区序列构建了带有His标签的甜菜M14品系半胱氨酸蛋白酶抑制剂基因的植物蛋白纯化表达载体,并将其转化烟草,获得T0代阳性转基因植株,为下一步利用融合蛋白的His标签从转基因烟草中大量纯化甜菜M14品系半胱氨酸蛋白酶抑制剂奠定了基础。

Abstract:It has been found that phytocystatin could improve crops ability of disease resistance and reduce the abiotic injury.The eukaryotic purification vector of the BvM14-cystatin had been constructed,which will express a His-Taged BvM14-Cystatin in plants.Moreover,some T0 generation transgenic tobaccos were selected for the purification of BvM14-Cystatin through affinity chromatography in a nickel column.This research established a solid foundation for purifying the BvM14-cystatin protein with the expression system of tobacco protein,and the characteristic of this protein is studied.
