甜瓜胚性愈伤组织诱导研究

摘    要:以甜瓜无菌苗4日龄子叶和1~4 mm2成熟种子切块为外植体,接种于不同培养基,在不同培养条件下,研究不同质量浓度的激素、不同暗培养时、不同类型外植体愈伤组织诱导的情况.研究结果表明,甜瓜成熟种子4日龄子叶外植体在加有2,4-D(1 mg/L),NAA(1 mg/L),6-BA(0.5mg/L)和蔗糖(3%)的MS培养基上先暗培养1周,再转移到光照条件25℃,16 h光周期下培养1周,最后转移到无任何激素的MS培养基中,胚性愈伤组织的诱导效果最好.

Abstract:Take 1 to 4mm2 sections of mature seeds and the 4 days old cotyledon as explants.The explants were inoculated on different media under different culture conditions to study the growth of somatic embryos in different hormone concentrations,incubation time and explants.The results showed that the 4 days old cotyledon of mature seeds of melon inoculated on 2,4-D(1 mg/L),NAA(1 mg/L),6-BA(0.5 mg/L)and glucose(3%)of the MS medium cultured under 1 week dark,and then transferred to light conditions 25℃,16 h photoperiod 1 week,transferred to MS medium without any hormone,induction of embryo callus were the best.
