珙桐休眠期种子高质量基因组DNA的提取及分析

摘    要:在对传统CTAB法进行改进的基础上,提取了沙藏珙桐休眠期去中果皮种子高质量基因组DNA,琼脂糖电泳及紫外分光光度分析,表明其OD260nm/OD280nm在1.8—2.0之间,用所提取的基因组DNA作为模板,S17为引物,进行RAPD扩增,获得了清晰的扩增图谱.

Abstract:Based on improving the traditional CTAB method,the high quality genome DNA was extracted from Davidia involucrata seed in dormant period.It indicated that the ratio of OD value at 260nm and 280nm was between 1.8 and 2.0 by agarose gel electrophoresis and UV spectrophotometry.Then,the extracted genome DNA was amplified by RAPD which used S17 as primer and the clear amplification patterns were obtained.
