L-甲硫氨酸高产菌株的紫外线诱变育种

摘    要:以一株具有M-海因水解酶系统产L-甲硫氨酸(L-Met)的芽孢杆菌MV0073为出发菌株,用含亮氨酸平板定向选育,经过6轮紫外诱变,筛选出系列L-Met高产突变株,其中菌株M604的L-Met产量最高.在相同培养条件下,M604的L-Met产量是出发菌株的3.14倍,且L-Met能以较高含量持续到发酵结束.M604连续传代6次,遗传性状比较稳定.

Abstract:UV mutagenesis was performed on Bacillus MV0073 which has L-5-thiomethyhydantoinase system. The mutants were selected directionally on the agar plate containing Leucine,and a series of new L-methionine-overproducing mutants was obtained by six UV mutagenesis One of the mutants M604 at the same cultured condition,the production of L-methionine by the strain was 3 14 times of that by original strain.The mutant M604 was generated for six times and showed that the L-methionine-overproducing capacity was stable.
