Abstract:Objective The study was to observe the effect of Curcumin inducing apoptosis in human hepatocellular carcinoma HepG2 cells, and to investigate its possible mechanism. Methods Experiments were divided into negative control group, 1 mg/ml curcumin group, 1.5 mg/ml curcumin group,2 mg/ml curcumin group. MTT assay was used to determine the proliferation inhibition of HepG2 ceils. The morphological changes of cells were observed by transmission microscope. The cell apoptosis was detected by flow cytometry (FCM). Western blot analysis was used to examine the expression of bcl-2 and survivin protein in each group after treated 24 h. Results A concentrations of Curcumin had a certain degree of growth inhibition on HepG2 ceils, and showed a dose- dependent and time-dependent manner. Apoptotic body could be observed under transmission microscope. FCM analysis demonstrated that the apoptosis rates of in each group were (4. 85 ± 0. 31 ) %, ( 8.25 ± 0.46 ) %, ( 11.44 ±0. 56 ) %, and ( 17.68±0. 98 ) % respectively. Western blot analysis demonstrated that the expression level of bcl-2 and survivin protein decreased with the increasing of Curcumin concentration. Conclusion Curcumin significantly inhibited the proliferation and induced cell apoptosis in HepG2 cells, and its mechanism may through down-regulating the expression of bel-2 and survivin.
