光谱法研究辛伐他汀与牛血清白蛋白的相互作用

摘    要:采用荧光光谱、圆二、傅立叶变换红外光谱及三维荧光光谱等分子光谱法研究了在生理条件下,辛伐他汀(Sire)与牛血清白蛋白(BSA)的结合作用.结果表明,Sire对BSA的猝灭方式为静态猝灭.根据F6rster非辐射能量转移理论,计算了Sim与BSA结合部位与色氨酸残基的距离r=1.8nm.利用标记药物进行了结合位点的定位,确定了Sim结合位置是BSA的siteI.由圆二、红外及三维光谱可知,Sim对BSA二级结构的改变主要在肛折叠区,在与Sire结合反应过程中,BSA的微环境与构象都发生了变化;同步荧光表明,Sim与BSA的作用部位主要在色氨酸(Trp)残基周围.

Abstract:Under the simulative human physiological condition, the interaction of the simvastatin (Sire) and the bovine serum albumin (BSA) was investigated by fluorescence spectra,Circular Dichroisrn and three-dimensional fluorescence spectra. The results showed that Sim could strongly quench the intrinsic fluorescence of BSA through a static quenching procedure. Based on the F6rster theory of non-radiative energy transfer,the distance between Sire and BAS was calculated to be 1.8 nm. In addition,the experi- ments suggested that the primary binding site for Sim was located at site I of BSA. According to the Circular Dichroism and three-dimensional fluorescence spectra,some micro-environmental and conforma- tional changes happened in the BAS molecules,and the changes of secondary structure were mainly in ]~- sheet of BSA. Synchronous fluorescence spectra indicated that Sirn and BSA mainly played roles in the residual group of tryptophan (Trp).
