摘    要:为探讨五味保肝丸对酒精性肝损伤小鼠的保护作用及其作用机制,将健康雄性昆明小鼠随机分为正常对照组、模型组、五味保肝丸低、中、高剂量组,每组10只进行实验。五味保肝丸组分别给予不同剂量五味保肝丸混悬液,正常对照组和模型组灌胃给予20 g/L羧甲基纤维素钠溶液,灌胃给药2 h后,除空白对照组外,给予56%白酒灌胃12 mL/kg,连续10 d,建立小鼠酒精性肝损伤模型。检测ALT、AST活性,肝微粒体CYP450含量以及CYP2E1蛋白的表达。研究结果表明:与模型组相比,五味保肝丸各剂量组可抑制急性酒精性肝损伤小鼠血清中ALT、AST含量的增加,增加CYP450含量,抑制CYP2E1蛋白的活化表达,其中,五味保肝丸中、高剂量组具有显著性差异(P<0.05)。五味保肝丸对小鼠酒精性肝损伤具有一定的保护作用,可能是通过保护细胞色素P450酶活性、抑制CYP2E1异常活化,发挥保肝降酶的作用。

Abstract:To investigate the protective effect and the mechanism of Wuwei Baoganwan(WBW) on alcohol-induced liver injury in mice, fifty healthy male KM mice were randomly divided into five groups: normal control (NC) group, model control (MC) group, high-,medium-,and low-dose WBW group, 10 mice in each group. To those in NC and MC group, 20 g/L CMC-Na Solution was given; those in high-,medium-,and low-dose WBW group were treated with WBW suspl. of 0.5, 0.25, 0.125 g/kg, respectively. The medication in all groups was given through intragastric administration, once a day for ten consecutive days. After 2 hours of administration every day, the mice of MC group and WBW groups were orally given 56% alcohol of 12 mL/kg to establish alcohol induced liver injury model. And to those in the NC group, equal dose of normal saline was given. The activity of ALT, AST, the content of CYP450 in liver microsome and the activity of CYP2E1 were respectively measured. Compared with the MC group, the ALT and AST levels were notably inhibited to increase in the three doses of WBW groups. The CYP450 content in the liver microsome were obviously increased. The activity of CYP2E1 was significantly decreased. Among them, there was significant difference between the middle and high dose of WBW groups(P<0.05).WBW shows protective effect on alcohol-induced liver injury in mice which might be related to its increasing the content of CYP450 and reducing the activity of CYP2E1.
