草珊瑚单细胞克隆的平板培养

摘    要:在草珊瑚细胞克隆的平板培养中,很多因素能影响其植板率。KT,2,4—D及NAA能提高草珊瑚细胞克隆的植板率,这三种激素对细胞生长的最佳搭配是KT0.4mg/1,2,4-D0.8mg/l,NAA0.2mg/l;草珊瑚细胞悬浮继代数不同,其植板率相差甚远,用悬浮培养第2代的细胞做材料最好;最适细胞克隆生长的培养基pH值为5.8;细胞植板密度低于每毫升500个细胞时,几乎没有克隆形成。

Abstract:Many factors could influence the plating efficiency (PE) of cell clone from Sarcandra glabra. Different concentrations of KT,2, 4 - D and NAA were able to influence the PE of a single cell of Sarcandra glabra.The best combination of these three hormones for the growth of a single cell was 0.4, 0. 8 and 0. 2 mg/1 respectively. The PE was obviously different from cells came from different generation of suspension subculture. The second generation of suspension culture cells had the best PE. The optimun plating medium PH for cells clone growth was 5. 8.There was few PE as the cells plating density was less then 500 cells/ml.
