| 摘 要： | 该文研究葡萄籽提取物(GSE)对前列腺癌PC-3细胞的生长抑制及凋亡作用,分析GSE诱导PC-3细胞凋亡的途径.采用四唑盐(MTT)比色法测定GSE对PC-3细胞的毒性作用;利用倒置显微镜、透射电镜观察GSE对PC-3细胞引起的形态学改变;碘化丙啶(PI)、吖啶橙(AO)/溴化乙锭(EB)双染法观察GSE诱导PC-3细胞凋亡的作用;Western blot探索细胞凋亡途径.结果表明,MTT法测定GSE能显著抑制PC-3细胞生长.倒置显微镜、透射电镜下可见细胞形态改变.PI染色显示,GSE将PC-3细胞周期阻滞在G0/G1期;GSE诱导PC-3细胞凋亡,AO/EB染色,荧光显微镜下可明显看到凋亡细胞、死亡细胞.Western blot显示GSE通过caspase途径诱导细胞凋亡.从细胞和亚细胞水平研究证实了GSE对人雄激素非依赖性PCaPC-3细胞具有毒性作用,GSE可通过线粒体途径诱导PC-3细胞凋亡和坏死,抑制细胞生长.

| Abstract: | This paper studies the effect of grape seed extract(GSE) on the proliferation and apoptosis of PC-3 cells.To analyze the pathway in which GSE induces apoptosis of PC-3 cells,MTT assay is applied to test the inhibitory effect of GSE on the proliferation of PC-3 cells.Inverted microscopy and transmission electron microscopy(TEM) are used to observe the morphologic changes in PC-3 cells.The effects of GSE on the apoptosis of PC-3 cells are studied by PI and AO/EB staining,and the pathway of apoptosis is invest... |
