摘    要:应用ISSR标记技术,对来自国内忍冬属(Lonicera Linn.)的20个居群进行遗传多样性和遗传结构的分析. 从100条ISSR随机引物中筛选出19条引物对20个忍冬居群进行扩增,共得到308条清晰的扩增位点,其中多态性位点278个,多态位点百分率(PPB)为90.26%. 遗传分化系数(GST)为0.5659,这显示20个居群的分子变异主要存在于居群间。AMOVA分析也显示居群间的遗传分化占到55.35%. 采用UPGMA法,根据ISSR-PCR结果进行聚类分析,得出20个忍冬居群遗传关系图,其种间关系与传统的形态学分类结果基本一致,但也有个别种的归属及种间关系稍有变化. 根据聚类图分析可知,一个称为“南江”的未知种和细毡毛忍冬(Lonicera similes Hemsl.)的三个居群聚在一起,表现出很高相似性,这说明它们之间的亲缘关系较为密切. “南江”可能是细毡毛忍冬的一个变种或栽培种. 本文为保存金银花种源植物的遗传多样性和进一步培育新品种提供了基础.

Abstract:In this study, inter-simple sequence repeats (ISSR) markers were employed to assess genetic diversity and population structure in twenty populations of Lonicera Linn, a source of anti-inflammatory herbal medicine. Using 19 screened primers, 308 discernible DNA fragments were generated of which 278 (90.26%) were polymorphic. The coefficient of gene differentiation (GST= 0.5659) showed that the total molecular variance of 20 populations was mainly existed among populations. AMOVA analysis showed that the genetic variation among the 20 populations was 55.35 %. Dendrogram based on ISSR-PCR markers was constructed by using UPGMA method, which almost agrees with results of traditional morphologic taxa, but hypotaxis status of few species was slightly changed. Based on the analyses, the so-called Nanjiang (an unknown species) and Lonicera similes Hemsl. were comprised a single cluster with a higher bootstrap support indicating Nanjiang would be a variation or subspecies of L. similes Hemsl. Our study provides a population-level genetic profile for further investigation and conservation of genetic diversity of Lonicera species related to Flos Lonicerae.
