不同高能饮食诱发胰岛素抵抗大鼠模型的比较

摘    要:目的分析高猪油日粮及高蔗糖日粮分别诱发胰岛素抵抗大鼠的血液生化指标差异,为此类模型的建立及实验研究提供参考。方法雄性SD大鼠随机分为3组,对照组给予普通日粮,高脂组给予高猪油含量的日粮,高糖组给予高蔗糖含量的日粮。喂养6周,每两周测定空腹血糖、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-c)、总胆固醇(TCH)、胰岛素,根据胰岛素敏感性指数(ISI)=ln1/(FPG×FINS)评定大鼠的胰岛素敏感性。结果 6周后,高脂、高糖组ISI显著低于对照组(P〈0.05);猪油组血清胰岛素显著高于对照组(P〈0.05)、血清HDL-c显著低于对照组(P〈0.05);高蔗糖组血糖、胰岛素均上升,但与对照组无统计学差异;而各组间血清总胆固醇、甘油三酯及体重无明显差异(P〉0.05)。结论高脂、高糖日粮均可诱发IR大鼠模型,高脂模型具有血清胰岛素显著升高、HDL-c显著降低的特点而高糖模型伴有较高的血糖、胰岛素。

Abstract:Objective To measure the biochemical parameters in blood of Insulin-resistance rat model induced by high-fat and high-carbohydrate diets.Methods Male SD rats were fed on standard laboratory chow,high-fat diet or high-carbohydrate diet respectively for 6 weeks.Serum glucose(GLU),insulin,triglyceride(TG),total cholesterol(TCH) and HDL-cholesterol were measured at fortnightly interval.The insulin sensitivity was estimated by ISI index,ISI=ln1/(FPG×FINS).Results Compared with control after 6 weeks,ISI index significantly decreased in both high-fat and high-carbohydrate groups(P<0.05),and serum insulin was significantly higher and HDL-cholesterol was significant lower in high-fat group(P<0.05).Serum GLU and insulin in high-carbohydrate group were higher than those in other groups.However there were mild difference in TG、GLU and body weight among the three groups.Conclusion Both high-fat and high-carbohydrate diets can induce insulin-resistant in rats.High-fat feeding results in significantly high serum insulin and low HDL-cholesterol,but high-carbohydrate treatment leads to high serum GLU and insulin.
