摘    要:以人前列腺癌PC3细胞作为研究对象,探讨了CCK-8法和MTT法的最佳实验条件。CCK-8的最佳检测波长为450nm,最佳检测时间为加入CCK-8试剂后4h,适宜细胞数量范围为8×10^2-1×10^5个。在检测抗肿瘤药物阿霉素(ADM)对PC3细胞的增殖影响实验中,发现CCK-8法测得数据的线性相关性优于MTT法,并且数据偏差较MTT法小。实验结果表明CCK-8法较MTT法检测的灵敏度高,准确性好,是一种优于MTT法的检测细胞增殖活性的方法。

Abstract:The optimal experiment conditions of CCK-8 and MTT in PC3 cell proliferation assays were investigated. For CCK,8 assay, the best condition is : 450 nm for detecting wave length, detected at 4 h after treatment, the number of PC3 cells is around 8 × 10^2 - 1 × 10^5. The antiproliferation action of ADM on PC3 were tested by CCK-8 and MTT. It was found that the result of CCK-8 method has better linear eorrelation fact, smaller deviation, higher sensitivity and accuracy. The conclusion is that CCK-8 method is superior to MTr method in cell proliferation assays.
