| 摘 要： | 通过密度梯度超速离心、脱脂从急性相胸水中分离得到含有血清淀粉样物质A（Serum AmyloidA,SAA）的高密度脂蛋白（HDL），再通过两次Sephacry1S-200层析纯化SAA蛋白。经SDS-PAGE电泳和Western Blotting检验证实得到的SAA蛋白纯度高，可作为抗原免疫动物制备抗体，方法可用于从胸水中大量制备SAA。

An Efficient Method for Purification of Serum Amyloid A (SAA) |

| Abstract: | A simple and efficient procedure was developed for purification of serum amyloid A (SAA) from acute_phase pleural liquid of human. The procedure included the preparation of high_density lipoprotein (HDL) by density_gradient ultrocentrifugation and two steps of gel filtration chromatography on Sepharcryl S_200. The first step of gel filtration chromatography was under the existence of 6 mol/L urea, and the second step was under the existence of 7 mol/L urea and 0.1% M_SH. The analysis of SDS_PAGE and Western Blotting showed that the obtained SAA protein could be used as antigen to immunize animals to produce antibody for the clinical usage. The developed procedure is applicable to large_scale preparation of SAA from pooled human pleural liquid. |
