藻胆蛋白表达条件优化的研究

摘    要:为了提高藻胆蛋白的表达含量,对本实验室构建的含pACYCDuet-ho1-pcyA、pETDuet-pecE-pecF、pCO-LADuet-pecA的E.coliBL21菌种的表达条件进行优化。通过单因素实验和正交实验L9(33)优化了蛋白的表达条件为:诱导表达温度为28℃,诱导表达时间为14h,诱导剂量为1.0mmol/L,藻胆蛋白最高表达含量为11.271mg/100mL.

Abstract:Objective The study is to optimize the culture conditions of recombinant phycobiliprotein expressed in E.coli BL 21.Through single factor and orthogonal design L9(33)experiments to make sure of the optimum conditions for culture condition.The results show that induce temperature 28℃,induce time 14h and inducer dose 1.0 mmol/L are the optimum conditions,and the yield of phycobiliproteins is 11.271 mg/100 mL.
