枯草芽孢杆菌TY7210菌株的ERIC-PCR快速鉴别

摘    要:以ER IC核心序列为引物,以2株枯草芽孢杆菌基因组DNA为模板,采用PCR对T aq DNA聚合酶用量、PCR退火温度及模板DNA浓度进行优化,建立了快速鉴别芽孢杆菌的ER IC-PCR方法,并对5株芽孢杆菌进行了分析.结果表明枯草芽孢杆菌TY 7210菌株与其他芽孢杆菌菌株具有不同的指纹图谱,尤其不同于枯草芽孢杆菌(ATCC 1.1849),为芽孢杆菌的快速鉴别奠定了基础.

Abstract:To set up a rapid identification method for Bacillus subtilis,the amplified system of ERIC-PCR was optimized,by changing the dosage of Taq DNA polymerase,annealing temperature and the concentration of the template DNA.The genomic DNA from two strains of Bacillus subtilis were used as the template to amplify by the primer of central sequence of ERIC and the fingerprints of five strains of Bacillus spp.were analysised by using optimized procedure of ERIC-PCR.The results showed that Bacillus subtilisTY_(7210) is different from others,especially Bacillus subtilis ATCC 1.1849.It contributed to the quick identification Bacillus spp.
