微波辅助萃取-高效液相色谱法测定苦丁茶中熊果酸的含量

摘    要:采用微波辅助萃取—高效液相色谱法测定了苦丁茶中熊果酸的含量.苦丁茶粉碎后以乙醇为溶剂,微波高火提取4 min,采用高效液相色谱法(HPLC)测定熊果酸的含量.色谱柱为InertsilC18柱(5μm,4.6×150 mm);流动相为甲醇/水(体积比90∶10),流速为0.8mL/min,柱温40℃,检测波长为210nm.结果表明熊果酸在0.0996~0.4980mg/mL范围内线性关系良好,平均回收率为100.8%,RSD=1.12%,苦丁茶中熊果酸的含量为14.55 mg/g.

Abstract:Using microwave extraction—High Performance Liquid Chromatographic(HPLC) method to detecte the content of ursolic acid in kudingcha.The kudingcha were crushed and were extracted by microwave-assisted.The microwave extraction method as follows: ethanol as solvent,microwave power of middle fire and extraction time of 4 minute.The analysis was carried out on a Inertsil C18 column(5 μm,4.6×150 mm)with a mobile phase of methanol/ H2O(V∶V=90∶10),at flow rate of 0.8 ml/min.The column temperature was set at 40℃ and the wave—length was 210 nm.The result indicated that the linear ranges was 0.099 6~0.498 0 mg/mL,the average recovery rate was 100.8% and the RSD was 1.12%(n=5).The content of ursolic acid in kudingcha was 14.55 mg/g.
