摘    要:目的 从细胞分子水平研究电压门控型钾通道亚型Kv1.5、Kv2.1在15-酮基二十碳四烯酸(15-ketoeicosatetretraenoic acid,15-KETE)致大鼠肺动脉收缩过程中的作用.方法 通过酶法分离、培养SD大鼠肺动脉平滑肌细胞(pulmonary arterial smooth cells,PASMCs),使用RT-PCR和Western blot技术观察15-KETE对大鼠PASMCs上Kv1.5、Kv2.1通道表达的影响.结果 (1)15-KETE下调大鼠PASMCs膜上Kv1.5通道mRNA及蛋白质的表达;(2) 15-KETE下调大鼠PASMCs膜上Kv2.1通道mRNA及蛋白质的表达.结论 Kv1.5、Kv2.1通道参与由15-KETE引起的缺氧肺动脉收缩(hypoxic pulmonary vasoconstriction,HPV).

Abstract:Objective To observe the effect of Kv1.5、Kv2.1 channels in rat pulmonary arterial smooth cells on the process of pulmonary vasoconstriction induced by 15-KETE.Method Western blot and PCR were used to identify the effect of 15-KETE on expression of Kv1.5、Kv2.1 channels in cultured rat pulmonary arterial smooth muscle cells (PASMCs).Result (1) The expressions of Kv1.5 mRNA and protein in rat PASMCs were significantly down-regulated by 15-KETE.(2)Theexpressions of Kv2.1 mRNA and protein in rat PASMCs were significantly down-regulated by 15-KETE.Conclusion Kv1.5、Kv2.1 channels are involved in vasoconstriction induced by 15-KETE in rat PA.
