秋水仙碱与DNA的相互作用

摘    要:
298.15K下,应用等温微量热法研究了抗肿瘤药物秋水仙碱(COL)与小牛胸腺DNA(ctDNA)结合作用,测定了药物与DNA分子的结合比、结合常数、结合焓变(△H°)、熵变(△S°)及吉布斯自由能变(△G°)等热力学参数.结合应用紫外-可见吸收光谱及荧光光谱研究了秋水仙碱与小牛胸腺DNA的分子间相互作用,探讨了药物对DNA分子构象的影响.

Abstract:
The interactions between the antitumour drug colchicine (ab.COL) and calf thymus DNA (ab.ctDNA) have been investigated with nano-Watt-scale isothermal titration calorimetry (ITC) at 298.15 K.Thermodynamic parameters such as the binding ratios of drug,binding constants,standard changes of enthalpy (△H°),Gibbs free energy (△G°) entropy (△S°) have been derived from the thermometric data.The influence of drug on molecular structure has been discussed according to UV absorption and fluorescence spectra.
