HPLC测定天麻素的含量和有关物质

目的建立测定天麻素原料药中天麻素的含量及有关物质的高效液相色谱法．方法采用Hypersil C18色谱柱，甲醇·0．01moL·L^-1磷酸二氢钠（8：92，磷酸调节pH至4．0±0．1）为流动相，检测波长为220nm，流速1．0mL·min^-1．结果天麻素在2．5～40．0ug·mL^-1范围内，峰面积与其浓度线性关系良好（r=0．9999），高、中、低3种浓度的平均加样回收率为99．7％～100．3％RSD为0．44％～0．82％，有关物质的限量为1．0％．结论本法准确、简便、快速，适用于天麻素原料药的质量控制．

Determination of Gastrodin and related substances in Gastrodin by HPLC

Objective： To establish an RP-HPLC for determination of Gastrodin and related substances in Gastrodin. Method： Gastrodin and related substances are determinated on HPLC instrument fixed with a Hypersil C18 column （250mm × 4.6mm,10 um） with UV detection at 220nm, and the mobile phase consists of phosphoric buffer（ 0.01moL · L^-1 NaH2PO4） and methyl alcohol （98：2, adjusted to pH4.0±0.1 with phosphoric acid）, the flow rate is 1.0 ml·min^-1. Result： The calibration curve is linear （r = 0.9999） within the range of 2.5 - 40.0 ug·mL^-1 for Gastrodin, the average recovery rate is from 99.7% to 100.3% for three different levels of the amount ofGastrodin, RSD is from 0.44% to 0.82%. The limit of related substances is 1.0%. Conclusion： This method is simple, quick, accurate and effective for testing related compounds in Gastrodin and for its contents determination. This method is suitable for the quality control for Gastrodin.
