狭叶薰衣草离体培养技术研究

摘    要:以狭叶薰衣草(Lavandula angustifolia Mill.)为材料,初步研究激素种类与水平对其愈伤组织诱导、不定芽增殖与不定根形成的影响.结果表明:狭叶薰衣草离体培养适宜的培养基分别为:诱导愈伤组织的培养基为MS+2,4-D 2.0 mg/L+6-BA 2.0 mg/L+蔗糖30 g/L+琼脂7 g/L;不定芽启动培养基为MS+6-BA 0.1mg/L~0.5mg/L+蔗糖30 g/L+琼脂7g/L;待小芽长至1~2 cm后将其转到1/2MS+IBA 0.5 mg/L+蔗糖30 g/L+琼脂7g/L培养基上生根,得到完整再生植株.

Abstract:In this experiment, using leaf or tip bud of Lavandula angustifolia as the explant, the effects of external hormone in medium on inducement of callus, multiplication of adventitious buds, and formation of adventitious roots were studied ; The results showed that medium for callus inducement should better be MS + 2, 2.0 mg/L of 4-D + 2.0 mg/L of 6-BA + 30g/L of sucrose + 7.0 g/L of agar. MS + 0.1-0.5mg/L of 6-BA + 30g/L of sucrose + 7g/L of agar was good for multiplication of adventitious buds. when the shoots grow to 1 ~2 cm high, they were cut from leaves, then transferred on 1/2MS+0.5 mg/L of IBA + 30g/L of sucrose + 7g/L of agar and developed into whole plants.
