摘    要:目的探讨肠道肿瘤患者与健康人群肠道菌群构成及基因组DNA的变化特征.方法分别采集肠癌患者与健康人晨起粪便,进行需氧和厌氧培养,鉴定菌株并进行菌落计数.采用试剂盒法提取粪便中肠道细菌总基因组DNA,进行琼脂糖凝胶电泳.结果肠癌组与健康对照组比较,肠杆菌数量增多,以大肠埃希菌数量显著,乳酸杆菌和双歧杆菌明显减少,厌氧菌和需氧菌的数量比例颠倒,两组数据比较差异均具有统计学意义(P0.05).粪便总基因组DNA经琼脂糖凝胶电泳在23 130 bp处有清晰、明亮的单一条带,且肠癌组与健康对照组均有相同大小的条带.结论肠癌组与健康对照组肠道菌群组成存在差异,肠道菌群总基因组DNA没有变化.

Abstract:Objective To explore differences in the composition of intestinal flora between the colorectal cancer patients and the healthy group in order to find an alternative method for the early prognosis of colorectal cancer. Method Early morning stool of the cancer patients and the healthy people were collected. Aerobic and anaerobic cultures were carried out,the strains were identified,and the bacterial colonies were count. Bacterial genomic DNA in feces was extraced from the colorectal cancer and the healthy group,respectively,and underwent agarose gel electrophoresis. Results Compared the cancer group with the control group, the amount of E. coli increased significantly,the amount of lactobacillus and bifidobacterium reduced significantly,and the proportional quantity of anaerobic bacteria and aerobic bacteria reversed. All the datum showed statistically significant (P<0. 05). There was a clear and bright single band at 23 130 bp on the agarose gel electrophoresis. Conclusion There were differences in intestinal flora composition between the cancer patients and the healthy control group, but their total genomic DNA was the same.
