青霉PT95菌株渗出液的产生及理化性质的研究

摘    要:实验观察发现,青霉PT95菌株渗出液的产生是由于菌丝互相挤压,紧密地缠绕在一起形成菌核,导致菌丝细胞液渗出,在菌落表面形成大量的渗出液.渗出液的物理和化学性质研究结果表明:渗出液的颜色随着培养时间的延长而逐渐变黄;渗出液的体积在第10天达到最大,然后迅速减少,第14天完全消失;干重随着培养时间的延长而减少.渗出液的pH值在5-6之间变化.渗出液中含有无机盐离子、氨、蛋白质、葡萄糖以及一些酶类.随着培养时间的延长,渗出液中的总蛋白、氨和无机盐离子(K^+、Na^+、Cl^-、P3^+和Mg^2+)的浓度都呈现明显下降趋势.只有Fe^2+浓度和酸性磷酸酶活性表现为逐渐增高的趋势,葡萄糖在渗出液中含量很低,丙氨酸氨基转移酶、门冬氨酸氨基转移酶活力变化不大.

Abstract:Penicillium sp.PT95 could form exudates on the colony surface during the development of sclerotia.In the zone where sclerotia formed,there was a compact mass of mycelia on which quantities exudates were present.Physiological and biochemical properties of exudates from Penicillium sp.PT95 were studied.The results showed that the exudates were pale yellow as the culture time was prolonged.At 10th day a lot of exudates were present,and at 13th day the exudates were fewer.The pH of exudates varied from 5 to 6.The exudates were found to contain cations,ammonia,protein,glucose and various enzymes.In aging cultures the dry matter weight,total protein,free ammonia,cations(K ,Na ,Cl-,P3 and Mg2 ) decreased whereas Fe2 and acid phosphatase in the exudates increased.
