三种常用方法保存临床常见菌种的比较

比较3种保存临床常见菌种常用方法.临床分离非苛养菌550株分别用半固体琼脂法、细菌湿种牛奶冻存法和纸片冻存法进行保存,比较保存1个月、12个月、36个月后3种方法的菌种存活率.半固体琼脂法、细菌湿种牛奶冻存法和纸片冻存法保存1个月时的菌株平均存活率分别为98.6%、100%和100%,36个月时的菌株平均存活率分别为45.1%、90.6%和86.1%.冻存法在长时间保存菌株时效果明显优于半固体琼脂法,3种方法对化脓性链球菌的保存效果不佳.结果表明,3种方法均能很好地保存临床常见菌,方法简便易行,各实验室可依具体要求选用.

Comparison of Three Methods for Common Clinical Bacterial Preservation

Three ordinary methods of preverving common clinical bacteria were compared.The semi-solid method(SSM),the milk freezing method(MFM),and the paper freezing method(PFM) were used to preserve 550 strains of non-fastidious bacteria from clinical isolates.Their bacterial survival rates(BSR) were compared respectively after bacterial preservation for 1,12,36 months.The results showed that the BSR of SSM,MFM,PFM were 98.6%,100% and 100% after preserving for 1 month,respectively.The BSR of SSM,MFM,PFM were 45.1%,90.6% and 86.1% after preserving for 36 months,respectively.The storage effect of freezing methods was better than that of SSM after long time preservation.But for three methods,the preservation of streptococcus was not good.As a result,the three methods are good for the common clinical isolates;they are simple and feasible,so any laboratory can select them according to its specific requirements.
