性激素是HPV16—DNA永生化人外宫颈上皮细胞的弱调节剂

摘    要:研究激素对HPV16-DNA永生化人外宫颈上皮细胞(HCE16/3细胞系)体外生长的影响。方法:使用[3H]-胸苷掺入试验,软琼脂糖集落试验和Northern印迹法分析了性激素对HCE16/3细胞的生长调节和病毒基因的表达。结果:在无类固醇血清无酚红的培养条件下,雌二醇和孕酮对HCE16/3细胞的生长无明显影响,而胰岛素则是HCE16/3细胞的生长刺激素。胰岛素的刺激细胞生长作用不能被性激素所增强;雌二醇也不能诱导HCE16/3细胞停泊独立生长。但Northern印迹分析显示性激素上调HCE16/3细胞病毒早期基因表达,该基因的表达可能与HPVDNA永生化细胞的生长有关。结论:HCE16/3细胞的生长仍然依赖于生长因子,性激素刺激病毒早期基因表达,要阐明性激素对人宫颈癌发生所起的作用,仍需做更多的工作

Abstract:Objective:To examine the effect of sex hormones on growth and gene expression of human papillomavirus (HPV) type 16 DNA-immortalized human cervical epithelial cells (HCE16/3 cells). Methods: The effect of sex hormones on growth and viral gene expression of HCE 16/3 cells were studied using -thymidine incorporation,soft agarose assay and Northern blot blot analysis. Results: The growth of HCE16/3 cells were found to be little affected by estradiol and progesterone, while insulin was a mitogen for HCE16/3 cells in phenol red-free medium with steroid-stripped serum.Furthermore,synergistic effects between insulin and hormones were not observed.Estradiol conld not induce growth of HCE16/3 cell line in soft agarose,too.In Northern blot analysis, however,hormones upregulated the HPV 16 early gene expression in HCE 16/3 cells,which is generally considered to require for the proliferation of HPV DNA-immortalized cells. Conclusions:These results suggest that the proliferation of HCE16/3 cells is still dependent on growth factors and sex hormones upregulate the HPV 16 erarly gene expression.More researches have to be done before elucidating the effects of sex hormones on human cervical carcinogenesis.
