木槿愈伤组织继代培养在不同培养基上的变化

摘    要:
木槿下胚轴在MS附加2,4-D(0.5mg/l)、激动素(0.1mg/l)和3%蔗糖的培养基(MS_1)上诱导出愈伤组织。将愈伤组织分别转移到不同培养基上,在特定培养基上可以发生根、芽和体细胞胚,并萌发成苗。组织切片观察了胚和器官的形态建成。最后,文中就激素、蔗糖浓度以及愈伤组织类型对形态建成的影响作了讨论。

Abstract:
Calli of shrubalthea (Hibiscus syriacus. L.) were induced from hypocotyl on MS medium supplemented with 2, 4--D (0. 5mg/l), kinetin (0. 1mg/l) and sucrose (3%) (MSl). Then the calli were transferred to four MS media. MS1, MS2, MS3, and MS0. After 3-4 weeks ,roots, shoots and somatic embryos were formed from certain calli cultured on given media. Somatic embryos matured and germinated on MS2 or MS0. Histo-cytological examination of calli revealed the embryo initiation and all stages of embryo development. Last of all, the effects of different hormones, suclose concentratiom amd callus type on the rmorphogenesis were discussed.
