摘    要:用发根农杆菌A4对荞麦的下胚轴和子叶进行离体转化,得到RiT-DNA表达的发状根。不同的培养环境对发状根的和生长有一定的影响,表现为发状根在液体培养基上比在固体培养基上生长更为迅速,且在培养基中含一定浓度的Cu^2+有助于发状根的形成。

Abstract:Cotyledon and hypocotyl of buckwheat were inoculated and cocultured with Agrobacterium rhizogenes A 4 strain .The transformed hairy roots was effected by different culture conditions.The hairy roots in liquid medium grew more faster than that in solid medium.The medium inculuding certain concentrated Cu 2 could promote the growth of hairy roots.Different explants and preculturing time showed certain influences to the transformation .The transformation frequency of hypocotyl was higher than that of cotyledon. Before the inoculation with Agrobacterium , a two day preculture of recipient explants on the phytohormone containing medium could obviously promote the transformation.Paper electrophoresis revealed that the hairy roots synthesized agropine and mannopine.
