摘    要:目的比较BHK-21细胞在3种生物材料溶液中黏附及增殖情况,探讨以类人胶原蛋白为基础制备组织工程材料的可行性。方法将BHK-21细胞分别种植于类人胶原蛋白、壳聚糖、透明质酸以及共混溶液中,在倒置显微镜下观察细胞的黏附和生长情况,并利用MTT方法检测种植后1~5 d细胞的增殖情况。结果BHK-21细胞种植12 h,类人胶原蛋白、壳聚糖以及两者共混溶液能显著促进BHK的黏附(P<0.01);BHK-21细胞在类人胶原蛋白溶液及类人胶原蛋白与其他生物材料共混溶液中功能活跃,维持其形态且能持续增殖,其中类人胶原蛋白/壳聚糖共混溶液促细胞增殖能力最强。结论类人胶原蛋白能够维持BHK细胞的生长与繁殖,在组织工程领域是一种极有潜力的组织工程材料。

Abstract:Aim To study the biocompatibility of materials for tissue engineering based on HLC and compare the attachment and proliferation of BHK cells cultured in three kinds of biomaterial solutions in vitro.Methods BHK cells were seeded into 96 plates to be cocultured with these solutions.Cell attachment and proliferation were observed using inverted light microscope.The cell proliferation was tracked by MTT method 1,2,3,4,5 days after seeding.Results BHK cells attachment could be stimulated by both HLC and the HLC-Chitosan blends significantly.Conclusion HLC can support the growth and stimulate the proliferation of BHK cells in vitro and thus,is a potential biomaterial in tissue engineering field.
