摘    要:采用蒽酮—硫酸法测定板栗多糖含量并对其测定条件进行优化。结果表明,蒽酮—硫酸法测定板栗多糖的适宜操作条件为:加入2g/L蒽酮—硫酸溶液4mL,在沸水浴中加热3min,自来水冷却40min后在10min内于波长600nm下读取吸光度。蒽酮—硫酸溶液需要现配现用。多糖含量控制在0.02~0.06g/L之间为宜。蒽酮—硫酸法测定板栗多糖含量的精密度实验RSD值为0.792%,平均加样回收率为107.2%,RSD值为4.6%。用本方法测定板栗中多糖含量为12.67%。

Abstract:The determination of chestnut polysaccharide content using anthrone-sulfuric acid method was studied and the operating conditions were optimized.The results showed that the optimal operating conditions were:4 mL of 0.2% anthrone-sulfuric acid solution,heated in boiling water bath for 3 min,cooled with city water for 40 min,read the absorbance under 600 nm wavelength in 10 min after cooling.The anthrone-sulfuric acid solution must be used quickly.Polysaccharide concentration between 0.02~0.06 g/L was the best for determination.The RSD in accuracy experiment was 0.792%,and the average recovery rate was 107.2% with RSD of 4.6%.The content of polysaccharide in chestnut determined by this method was 12.67%.
