皱纹盘鲍组织培养研究

摘    要:用MEM培养液对皱纹盘鲍的血细胞、鳃、外套膜、肾脏和足肌进行了组织培养研究,血细胞培养1h后即贴壁生长,从形态上可分为成纤维样细胞和圆形细胞,但2d后即脱落,鳃细胞呈圆形,增殖迅速,容易获得传代细胞,其中两株现已分别传至22代和30代,肾细胞呈上皮样,生长缓慢,已传至第3代、外套膜和足肌则未获传代细胞。

Abstract:Haemocytes, gill, mantle, kidney and foot muscle from Haliotis discus hannai Ino are cultured in vitro with MEM medium. After 1 hour incubation, the haemocytes adhere on the glass and multiplied. The haemocytes are distinguished into the fibroblast?like cells and round cells based on their morphology. But the cells detach from the surface of the plate after 2 day incubation. The gill cells, which are round, multiplied rapidly. The subcultures are easily set up from the gill. Two strains have been subcultured up to 20 and 30 generations respectively now. The kidney cells, which are epithelial?like, multiplied slowly and have been subcultured up to 3 generations now. The subculture from the mantle and the foot muscle is not set up.
