电镜生物样品预冷过程的理论分析与实验研究

本文从传热学角度对生物样品在浸入冷却剂之前的预冷过程进行了定量分析和实验研究。测量了冷却剂上方冷气层中温度的静态分布规律,研究了气层厚度、沉浸速度、样品直径和样品初始温度等因素对样品预冷过程的影响,提出了避免或减轻预冷对生物样品低温固定质量影响的措施。最后,定性地研究了预冷对电镜生物样品超微结构的影响。

THEORETICAL AND EXPERIMENTAL RESEARCHES ON THE PRECOOLING PROCESS OF BIOSAMPLES FOR ELECTRON MICROSCOPY

Both theoretical and experimental thermal quantitaive analyses of the precooling process of the sample before being plunged into the cryogen are carried out. The static temperature profiles in the cold gas layer above the cryogen is measured. The effects on the precooling process of the thickness of the cold gas layer, the plunge velocity, the radius of the samples and the initial temperature are investigated. The methods to avoid or relieve the harmful effects of precooling are proposed. In point of bio-electron-microscopy ultrastructure, the effect of the specimen precooling on cryofixation quility is also discussed.
