鱇浪白鱼ISSR-PCR最佳退火温度的筛选

摘    要:通过实验测定获得了鱇浪白鱼18条ISSR引物的最佳退火温度,发现不同引物间的差异较大,最高退火温度的引物825,为61.5℃,最低退火温度的引物876,为47.9℃,二者相差达到13.6℃.所有18条引物最佳退火温度的变异系数7.59%.理论推算与实际测量的最佳退火温度存在较大的差异,相关分析表明二者相关系数较低,仅为0.23.由此可见,理论推测的退火温度与实测值间不具有明显的相关性.

Abstract:The optimum annealing temperature of 18 ISSR primers was obtained in Anabarilius grahami. Obvious difference existed among these primers. The highest annealing temperature is 61.5 ℃ attributed to primer 825, which is higher 13.6 ℃ than the lowest annealing temperature which is 47.9℃ attributed to primer 876. The variance coefficient amounted to 7.59%. There was large difference of the annealing temperature between experiment and theory. The correlation coefficient is only 0.23. These results show that there is no obvious correlation relationship of the annealing temperature between theory and experiment.
