摘    要:目的观察与分析冠脉结扎法与给药法制做的大鼠心肌缺血模型。方法缝扎大鼠冠脉左前降支,于左室前外侧壁形成缺血区域,约占左室壁面积的20%~50%;经给药造模,在皮下一次性多点注射盐酸异丙肾上腺素(Iso)。结果结扎18例动物制作模型,存活11只,病理检查证实50%有明确的心肌缺血,心功能下降;而给药组18只动物模型,存活17只,病理检查证实80%有明确的心肌缺血。结论给药方法制作更简单可行,动物存活率高;而结扎模型欠稳定,需标本量较大。

Abstract:Objective To compare the experimental rat models of myocardial ischemia induced by coronary artery ligature and administration of isoproterenol hydrochloride ( ISO). Methods For coronary artery ligature, the left anterior descending coronary arteries of rats were ligatured, then the ischemic areas about 20% to 50% of the left ventricular wall were observed in the left ventricular lateral wall; for administration of ISO, one-time multi- subcutaneous injection of isoproterenol hydroehloride was performed to each rat. Results Eleven of the 18 myocardial ischemia rats induced by coronary artery ligature survived, and pathological examination revealed clear myocardial ischemia and reduced cardiac function in 50% of the whole rats; in the IS0-induced group, 17 of the 18 model rats survived, and pathological examination revealed a clear lack of blood supply to heart in 80% of the rats. Conclusion Induction of rat myocardial ischemia model by administration of ISO is simple and practical, and results in high survival rate in rats.
